چکیده
Purpose: Monoclonal gammopathy of undetermined significance (MGUS) is a plasma cell dyscrasia. It known that MGUS has an increased risk progression to multiple myeloma (MM), and also prepares the ground for diseases such as Waldenstrom macroglobulinemia (WM), non-Hodgkin lymphoma, chronic lymphocytic leukemia (CLL). Our study aimed evaluate whether some important p53 pathway genes differ in terms expression healthy individuals.
Materials methods: For study, bone marrow was collected from 8 individuals diagnosed with MGUS, RNA samples were isolated. Expression levels various involved compared using RT2-profiler PCR array. β-actin housekeeping gene level used normalization. The Pearson Correlation analysis Receiver Operating Characteristic (ROC) conducted.
Results: Among whose examined this it determined only GADD45A decreased significantly control group (p=0.027). correlation data showed highly correlated 12 other (APAF1, CDK4, PCNA, BAX, CDKN2A, CASP9, CHECK2, MDM2, RB1, P53, BCL2, CHEK1) (r>0.7). In addition, according ROC analysis, detected have strong discrimination power between (AUC=0.797 p=0.015).
Conclusion: may be useful new biomarker detect MM.

متن کاملGADD45a physically and functionally interacts with TET1
DNA demethylation plays a central role during development and in adult physiology. Different mechanisms of active DNA demethylation have been established. For example, Growth Arrest and DNA Damage 45-(GADD45) and Ten-Eleven-Translocation (TET) proteins act in active DNA demethylation but their functional relationship is unresolved. Here we show that GADD45a physically interacts--and functionall...
متن کاملGADD45A Does Not Promote DNA Demethylation
Although DNA methylation patterns in somatic cells are thought to be relatively stable, they undergo dramatic changes during embryonic development, gametogenesis, and during malignant transformation. The enzymology of DNA methyltransferases is well understood, but the mechanism that removes methylated cytosines from DNA (active DNA demethylation) has remained enigmatic. Recently, a role of the ...
متن کاملGadd45a promotes DNA demethylation through TDG
Growth arrest and DNA-damage-inducible protein 45 (Gadd45) family members have been implicated in DNA demethylation in vertebrates. However, it remained unclear how they contribute to the demethylation process. Here, we demonstrate that Gadd45a promotes active DNA demethylation through thymine DNA glycosylase (TDG) which has recently been shown to excise 5-formylcytosine (5fC) and 5-carboxylcyt...
متن کاملIng1 functions in DNA demethylation by directing Gadd45a to H3K4me3.
Active DNA demethylation regulates epigenetic gene activation in numerous processes, but how the target site specificity of DNA demethylation is determined and what factors are involved are still poorly understood. Here we show that the tumor suppressor inhibitor of growth protein 1 (Ing1) is required for targeting active DNA demethylation. Ing1 functions by recruiting the regulator of DNA deme...
